Abstract. Angiotensin II stimulates the hepatic synthesis and secretion of angiotensinogen, the substrate of renin. In the present study performed on freshly isolated rat hepatocytes we demonstrate that this effect of angiotensin II . The role of the liver in synthesis of coagulation factors was studied by perfusion of isolated, intact rat liver. Direct evidence for synthesis of prothrombin and Factors V, VII and X by the liver was obtained. Inhibition of synthesis of prothrombin, Factor VII and Factor X by Coumadin and reversal of inhibition by vitamin K1 were by: The hepatic pattern of synthesis of the apolipoprotein A-I isoforms has been analyzed in the rat. After isolated livers were perfused with defibrinated rat blood and [3 H]leucine, the radioactivity associated with apolipoprotein A-I and other apolipoproteins was determined following two-dimensional gel electrophoresis of the perfusate d Cited by: GAA formation in rat is not detected in liver homogenates 2 or in isolated perfused livers 3. Therefore, creatine synthesis in rat liver is regulated by the activity of transamidinase in the kidney 4. However, recent improvements in GAA analysis reveal that the blood level is .
Vitamin K 1 induced the formation of prothrombin by the isolated perfused rat liver when administered in response was blocked by puromycin. When l-leucine-l-C 14 was present in the medium at the time vitamin K 1 was added, the amino acid was incorporated into the newly synthesized prothrombin which was precipitated by specific antibody. It would appear from these data that vitamin K. SARCIONE EJ. Sunthesis of alphal-acid glycoprotein by the isolated perfused rat liver. Arch Biochem Biophys. Bile acid processing in the long-term, bile duct obstructed rat liver was studied ex vivo. Twenty four and 72 h, respectively, after bile duct obstruction the isolated liver was perfused with. Isolated hepatocytes were prepared from rat liver by collagenase perfusion and density gradient centrifugation. The hepatocyte preparation released angiotensinogen at a basal rate of pmol/g. The production of plasma proteins, prothrombin and Factor VII, essential to the blood clotting process, is dependent on the action of vitamin K. In intact, vitamin K deficient rats, actinomycin D did not block the increase induced by vitamin K in plasma prothrombin and the clotting defect was restored to normal within the 6-h period. When the experiment was repeated with the addition of. In a previous study using isolated rat liver mitochondria, we observed that IMD inhibited the activity of F o F 1-ATP synthase. The aim of this study was to evaluate the effects of IMD on rat isolated hepatocytes and perfused rat liver, besides the influence of its biotransformation on the toxicological potential. The toxic effect of fluoroacetate was studied in a metabolic experiment on intact isolated rat heart. After the heart was perfused with mM fluoroacetate, the measured rate of glucose uptake and glycolysis decreased and glucosephosphate and fructosephosphate accumulated. An examination of the citric acid cycle intermediates indicated that their concentrations were below normal except.
